Un’alternativa all’uso del PVP (poli vinil pirrolidone) per la selezione degli spermatozoi da utilizzare al momento della ICSI è quella dell’acido ialuronico. L’uso del PVP permette di rallentare i movimenti dello spermatozo e di osservare lo spermatozoo morfologicamente migliore per il suo utilizzo nella microiniezione tenendo presente, altresì che tale sostanza potrebbe avere effetti tossici sull’ovocita.
Se si usa l’acido ialuronico è possibile effettuare una migliore selezione, in quanto nella fecondazione naturale l’acido Ialuronico è presente sulla superfice degli ovociti e, gli spermatozoi al momento della fecondazione devono possedere dei recettori per l’acido ialuronico affinchè vi sia il legame con l’ovocita e quindi la penetrazione nel suo interno. Affinchè avvenga questo colloquio recettoriale è necessario che lo spermatozoo abbia caratteri di normalità del suo DNA e dei cromosomi: il DNA deve essere integro e senza frammentazioni. Nel caso di PMA : al momento della ICSI, si crea un contatto tra gli spermatozoi da microiniettare negli ovociti e l’acido ialuronico che seleziona in un modo facilmente osservabile gli spermatozoi con DNA non danneggiato che possono quindi essere bloccati e iniettati singolarmente in un ovocita maturo (MII). L’uso dell’acido ialuronico, quindi permette di effettuare una ICSI che non utilizza il PVP ed evita i sui possibili effetti tossici sull’ovocita e nello stesso tempo permette una selezione di spermatozoi con DNA non frammentato. Nell’ambito del MASTER di II livello di Biotecnologie della Riproduzione che si tiene presso il dipartimento di Scienze per la Vita della II Università di Napoli, sede di Caserta si sta sperimentando tale tecnica usando modelli animali in un ambito collaborativo dove sono coinvolte strutture della facoltà che studiano la frammentazione del DNA degli spermatozoi. Un work in progress per dar modo agli studenti del Master di II livello di apprendere tale nuova tecnologia.
